- You should aim to achieve at least a Signal to Background ratio of 5 in your image (The structures of interest should be at least 5 times brighter than background e.g. Signal 500 / Background 100)

- Next, move to the Acquisition Mode window; Select as small ROI as needed for your sample (reduces file size)
- In Noise Filter click "On - TIRF or WF"
- Choose Number Phases: 13 phases provide max resolution for optimal SIM, 9 phases provide less resolution but are faster (therefore also less photobleaching)
